Method development for chromatography is not an exact science.

Method development for chromatogra-phy is the process of determining what conditions are most appropriate for the analysis required. The strategy that chromatographists apply to develop a method, is generally determined by three variables. First of all, accurate knowledge of target compound chemis-try is necessary to determine which se-paration technique, i.e. gas chromato-grapy (GC) or liquid chromatography (LC), has the most change of success.


In addition, the concentration level at which the target compound(s) need to be determined is important as well. Low concentration levels generally require a dedicated sample preparation approach, albeit in combination with a specific detector, such as a mass spectrometer (MS). Last but not least, the overall complexity of the sample matrix has to be taken into account to decide
on sample preparation approach, injection- and detection technique.
Method development
Chromatography
has never been an exact science and fortunately it still is not. There are seve-ral ways to come to a chromatographic method that can be used in practice. Most of the time, the final method is function of both the experience and creativity of the chromatographer and the availability of instruments/techniques in the laboratory.
Instrumentation
In practice, method development for any analytical technique starts with a survey of available literature. Depending on the complexity of the analytical question, this survey can be very short.
A smart query that uses the right keywords in Google can be sufficient to gain the necessary in-sights to start the initial experiments. If necessary, more detailed information can be found using dedicated websites such as Scirus, Pubmed, Chemweb and ScienceDirect.
